چکیده:
مقدمه: امروزه از سویا به سبب خصوصیات کاربردی و تغذیهای موجود در پروتئینهای آن در محصولات گوشتی استفاده میگردد. با این وجود سویا و مشتقات آنبر اساساستاندارد ملی ایران به عنوان مواد حساسیتزای شدید برای برخی افراد شناخته شدهاند که باید از قوانین برچسبگذاری تبعیت کنند. بنابراین هدف این پژوهشتوسعه یک آزمون PCR برای شناسایی سویا به عنوان یک ماده آلرژن پنهان در محصولات گوشتی میباشد.
مواد و روشها: در این تحقیق، ابتدا نمونههای سویا و محصولات گوشتی تجاری تهیه و DNA هر یک با استفاده از کیت MBST، استخراج و خالصسازی گردید. پرایمر اختصاصی سویا طراحی و سفارش داده شد. سپس شرایط آزمون PCR برای تکثیر قطعه 118 جفت بازی متعلق به سویا، بهینهسازی و محصولات PCR، بوسیله ژل آگارز 5/1% الکتروفورز گردید. در نهایت کارایی این تکنیک برای شناسایی سویا در محصولات گوشتی تجاری مورد ارزیابی قرار گرفت و نتایج آزمون PCR بکار رفته با اظهارات برچسب محصولات گوشتی مقایسه شد.
یافتهها: تحت شرایط بهینهسازی شده، قطعه DNA با طول 118جفت بازی برای سویا تکثیر یافت. سپس تکنیک توسعه یافته به طور موفقیتآمیزی برای شناسایی سویا در محصولات گوشتی تجاری مورد استفاده قرار گرفت، نتایج نشان داد که 100% محصولات سوسیس و ناگت مرغ ، 33% محصولات کالباس، 60% محصولات همبرگر، 75% محصولات کبابلقمه باند 118 جفت بازی متعلق به سویا را ظاهر ساختند.
نتیجهگیری: نتایج نشان داد آزمون PCR توسعه یافته در مطالعه حاضر، روشی سریع، حساس، ساده، قابل اعتماد و مقرون به صرفه برای شناسایی پروتئین گیاهی سویا به عنوان یک ماده حساسیتزا در محصولات گوشتی تجاری خام و حرارت دیده میباشد..
Introduction: Soya is used in meat products due to its functional and nutritional properties.
Soya is also known as an intense allergen substance for some people according to the Institute
of Standards and Industrial Research of Iran, therefore its presence must be mentioned on the
label of the products. The purpose of this study is to develop a PCR assay for identifying soy
protein as a hidden allergen substance in meat products.
Materials and Methods: In this study, soya and meat products samples were prepared. The
extraction of DNA from samples were carried out according to the instruction provided by
MBST kit. The specific primer pairs of soya were designed and ordered. The PCR conditions
were standardized to obtain the desired amplicon of 118bp for soya. Finally the PCR products
were electrophoresed in 1.5% agarose gels. This method was used for detection of soya in
commercial meat products and the results were compared with their labeling statements.
Results: In optimized PCR condition, DNA fragment of 118bp for soya was amplified. This
developed technique was successfully evaluated for detection of soya in meat products. The
results showed that 100% of sausages and chicken nuggets, 33.3% of cold cuts, 60% of
hamburgers and 75% of Loghme Kebab contained soya.
Conclusion: The results indicated that PCR assay developed in this study was a rapid, sensitive, simple, reliable and commodious method that might be used for the detection of
soya in raw and heat treated meat products.
